Mouse NEI endonuclease VIII-like protein 3 (NEIL3) is a 77 kDa DNA glycosylase whose N-terminal helix-two-turn-helix motif pinches ring-opened purines, while its C-terminal zincless finger threads through the duplex to flip the damaged base. A solvent-exposed β-hairpin (aa 210-225) frames the catalytic lysine, presenting a conformational epitope unique among Fpg/Nei family members. Polyclonal antibodies directed against this hairpin trace NEIL3 in S-phase nuclei, mitochondrial matrix, and testicular germ cells, yielding crisp nuclear foci and single-band clarity on immunoblots without cross-reacting with NEIL1 or NEIL2.
