Mouse interleukin-35 (IL-35) is a 48 kDa heterodimeric IL-12 family cytokine composed of p35 and EBI3 subunits whose composite grooves engage gp130/IL-12Rβ2, delivering inhibitory cues to T cells and macrophages. A solvent-exposed 14-aa "KVE" loop (aa 85-98) at the p35 elbow region folds into a β-hairpin unique among mouse IL-12 members, presenting a conformational epitope preserved after secretion and fixation. Affinity-purified antibodies targeting this loop trace native IL-35 in regulatory T-cell supernatants, splenic follicles, and lamina propria, delivering crisp extracellular deposition and single-band clarity on immunoblots with no cross-reaction to IL-12 or IL-27.
