Target Information
Rat Macrophage Expressed Gene 1 Protein (MPEG1) is a pore-forming, type I transmembrane glycoprotein that belongs to the MACPF superfamily. It is constitutively and inducibly expressed in macrophages and dendritic cells, where it participates in both innate and adaptive immunity. Upon acidification of phagosomes, MPEG1 oligomerizes and inserts amphipathic β-hairpins into bacterial or endosomal membranes, creating transmembrane pores that facilitate antigen cross-presentation and intracellular killing of a broad spectrum of bacteria. Proteolytic processing by trypsin and asparagine endopeptidase, together with pH-dependent conformational changes, are required for pore assembly and antibacterial activity. Antibodies against MPEG1 enable precise subcellular localization and functional analysis of this macrophage-specific defense molecule in rat models .
