Mouse contactin-associated protein-like 4 (CNTNAP4) is a 150 kDa neurexin-family transmembrane scaffold whose laminin-G and EGF repeats bridge juxtaparanodal Kv1 channels to Caspr2, clustering at internodal axoglial junctions. A solvent-exposed 18-aa linker (aa 730-747) between discoidin and F5/8 modules folds into a β-hairpin unique among CNTNAP paralogs, offering a fixation-stable epitope. Polyclonal antibodies targeting this loop trace native CNTNAP4 in hippocampal trisynaptic circuits, optic nerve, and myelinated sciatic axons, yielding crisp juxtaparanodal doublets and single-band specificity on immunoblots with no cross-reaction to CNTNAP1 or CNTNAP2.
