Target Information
Mouse glypican-3 is a 66 kDa GPI-anchored heparan-sulfate proteoglycan whose N-terminal globular core and three HS attachment sites sequester Wnts and BMPs, sculpting growth-factor gradients at the cell surface. A solvent-exposed 16-aa "KVE" loop (aa 250-265) between the core and first HS domain folds into a β-hairpin unique among mouse glypicans, presenting a conformational epitope preserved after heparitinase treatment and fixation. Affinity-purified antibodies targeting this loop trace native glypican-3 in fetal liver, placental labyrinth, and limb-bud ectoderm, delivering crisp membrane rings and single-band clarity on immunoblots with no cross-reaction to glypican-1 or glypican-4.
