Target Information

Mouse neurofibromin 2 (NF2, merlin) is a 68 kDa FERM-domain protein whose N-terminal cloverleaf clamps plasma-membrane PIP2 while an α-helical rod folds back to auto-inhibit downstream Ras-PI3K signaling. A solvent-exposed 18-residue "EKL" loop (aa 185-202) between FERM subdomains B and C presents a β-hairpin unique among ERM/NF isoforms, offering a conformational epitope preserved after paraformaldehyde fixation. Affinity-purified antibodies targeting this loop trace native NF2 in meningeal lining, Schwann-cell paranodes, and renal tubule membranes, delivering crisp cortical rings and single-band clarity on immunoblots with no cross-reaction to ezrin, radixin, or moesin.
