Target Information

Rat cytochrome P450 27B1 is a 56 kDa inner-mitochondrial enzyme whose heme-thiolate core converts 25-hydroxyvitamin D to the active 1,25-dihydroxy form, steering calcium and phosphate homeostasis. A solvent-exposed 15-aa "KVE" loop (aa 220-234) between helices F and G folds into a β-hairpin unique among rat P450s, presenting a conformational epitope preserved after paraformaldehyde fixation. Affinity-purified antibodies targeting this loop trace native CYP27B1 in renal proximal tubules, placental labyrinth, and osteoblastic layers, delivering crisp mitochondrial reticula and single-band clarity on immunoblots with no cross-reaction to CYP24A1 or CYP27A1.
