Rat brain finger protein (RNF112, Znf179) is a 65 kDa RING-B-box-coiled-coil protein whose C3HC4 RING and two B-box domains ubiquitinate synaptic substrates, shaping neuronal differentiation and survival. A solvent-exposed 16-aa "KVE" loop (aa 240-255) between the second B-box and the coiled-coil folds into a β-hairpin unique among rat RNF proteins, presenting a conformational epitope preserved after fixation. Affinity-purified antibodies targeting this loop trace native RNF112 in cortical neurons, hippocampal mossy fibers, and cerebellar granule cells, delivering crisp nuclear and cytoplasmic puncta with single-band clarity on immunoblots and no cross-reaction to other RING finger proteins.
