Target Information

Mouse diacylglycerol kinase alpha (DGKα) is a 82 kDa lipid kinase whose tandem EF-hands sense Ca²⁺ and whose catalytic loop converts DAG to phosphatidic acid, terminating PKC signals. A solvent-exposed 18-aa "KAL" linker (aa 367-384) between the recoverin homology and catalytic domains adopts a β-hairpin unique among DGK isoforms, presenting a fixation-stable epitope absent from DGKζ or DGKθ. Affinity-purified antibodies targeting this linker trace native DGKα in T-cell membranes, neuronal growth cones, and mast-cell cytoplasm, yielding crisp punctate rings and single-band clarity on immunoblots with no cross-reaction to other DGK family members.
