Mouse coagulation factor VIII (FVIII) is a 265-kDa glycoprotein synthesized in hepatocytes and endothelial cells, circulating as a non-covalent complex with von Willebrand factor (vWF) in mouse plasma. It functions as a critical cofactor in the intrinsic coagulation pathway, activating factor X by forming a complex with factor IXa and calcium ions on phospholipid surfaces. This step amplifies thrombin generation, enabling fibrin clot formation and hemostasis. Essential for murine vascular integrity, its deficiency or dysfunction causes hemophilia A, characterized by spontaneous bleeding and impaired clotting. As a central mediator of physiological hemostasis, mouse FVIII is a key target for studying coagulation biology, hemophilia, and anti-bleeding therapeutic development.
