Rat nuclear factor, erythroid-derived 2-like 2 (NFE2L2/Nrf2) is a 68 kDa leucine-zipper transcription factor whose N-terminal Neh2 domain locks onto Keap1 until redox perturbation releases its DNA-binding CNC-bZIP module. A solvent-exposed hinge peptide (aa 16-30) harbors the critical DLG and ETGE motifs, providing a conformation-specific epitope present only in the free, active form. Polyclonal antibodies directed against this hinge region trace nuclear translocation in hepatocytes, alveolar macrophages, and renal tubules, distinguishing full-length Nrf2 from smaller proteolytic fragments or homologous small Maf proteins with single-band clarity on immunoblots and crisp nuclear staining in formalin-fixed tissue.
