Target Information
Mouse autophagy-related protein 7 (Atg7) is a 78 kDa E1-like enzyme that orchestrates the lipidation cascade essential for autophagosome expansion. Its N-terminal adenylation domain activates Atg12 and LC3 in succession, while the C-terminal ubiquitin-fold recruits the Atg10 and Atg3 E2 enzymes to catalyze two parallel conjugation pathways. A flexible linker between residues 400-450 swings the catalytic cysteine between donor and acceptor sites, enabling sequential transfer without substrate release. Polyclonal antibodies raised against the unique linker region detect both cytosolic and membrane-bound Atg7, distinguishing full-length from calpain-cleaved fragments in tissue lysates and immunofluorescence assays.
