Target Information
Protein-glutamine gamma-glutamyltransferase K (TGM1) facilitates the cross-linking of proteins and the attachment of polyamines to proteins. It is crucial for cross-linking epidermal proteins during the formation of the stratum corneum and plays a role in cell proliferation. In mice, the loss of the TGM1 gene results in severe skin barrier defects and neonatal death. This condition is characterized by poor keratinization, failure in assembling the cell envelope, and dissociation of the lipid bilayer. These defects prevent mice from surviving after birth because the compromised skin barrier function leaves them unable to adapt to the external environment. Additionally, the absence of TGM1 can lead to erythrodermic skin lesions.
