Rat mucin 5 subtype b (Muc5b) is a 600 kDa secreted gel-forming mucin whose N-terminal D-domains polymerize into disulfide-linked networks while central PTS repeats are densely O-glycosylated, creating the viscoelastic blanket of respiratory and gastrointestinal tracts. A solvent-exposed 18-aa "KVE" loop (aa 1,220-1,237) between D2 and D3 folds into a β-hairpin unique among rat mucins, presenting a conformational epitope preserved after reduction and fixation. Affinity-purified antibodies targeting this loop trace native Muc5b in tracheal goblet cells, submandibular glands, and gastric foveolae, delivering crisp apical granules and single-band clarity on immunoblots with no cross-reaction to Muc5ac or Muc2.
