Expressed on mast cells, macrophages, dendritic cells, and certain T-cell subsets, it binds fibronectin and MHC class I ligands, recruiting SHP-1/SHP-2 phosphatases to dampen FcγR, MAPK, and NF-κB signaling. This inhibition limits cytokine release, T-cell proliferation, and osteoclastogenesis, thereby moderating innate and adaptive immune responses. Constitutive yet inducible expression in lung, liver, and spleen makes LILRB4 a reliable marker for immune checkpoint studies.
