Rat Deoxyribonuclease I-like Protein 2 (DNASE1L2) is a secreted endonuclease that cleaves double-stranded DNA into 3'-hydroxyl oligonucleotides in a magnesium-dependent manner. It is constitutively expressed in stratified squamous epithelia, where it facilitates degradation of nuclear DNA during terminal differentiation of keratinocytes. The enzyme is stored in lamellar bodies and released into extracellular spaces, contributing to chromatin breakdown and barrier formation. Antibodies to DNASE1L2 enable specific detection of DNA fragmentation during epidermal maturation and wound healing in rat skin models without cross-reactivity to DNASE1 or DNASE2.
