Target Information
Rat axis inhibition protein 1 (Axin1) is an 86 kDa scaffold whose N-terminal RGS domain captures APC while a C-terminal DIX segment polymerizes with Dishevelled, nucleating the β-catenin destruction complex. A solvent-exposed 16-aa "KVE" loop (aa 380-395) between GSK3β- and β-catenin-binding sites folds into a β-hairpin unique among rat Axin paralogs, presenting a conformational epitope preserved after phosphorylation and fixation. Antibodies targeting this loop trace native Axin1 in hepatic cytoplasm, colonic crypts, and neuronal growth cones, delivering crisp punctate staining and single-band clarity on immunoblots with no cross-reaction to Axin2.
