Mouse Claudin 5 is a 22 kDa tetraspan tight-junction protein that seals the paracellular space of endothelial and epithelial cells, creating a high-resistance barrier selective for small ions. It is concentrated in brain microvessels, lung alveoli, and kidney distal nephron, where it limits solute flux and maintains tissue-specific microenvironments. Claudin 5 expression is dynamically regulated by oxygen, inflammatory cytokines, and developmental cues, controlling permeability without altering junction architecture. As the principal determinant of blood-brain barrier tightness, it is a key target for studies of neurovascular integrity, edema, and metastasis.
