Target Information
Rat uroplakin 3a (UPK3A) is a 34 kDa tetraspanin whose two extracellular loops and a conserved N-glycan assemble into urothelial plaque particles, forming the permeability barrier of the bladder apical membrane. A solvent-exposed 14-aa "KVE" loop (aa 85-98) between the first and second transmembrane spans folds into a β-hairpin unique among rat uroplakins, presenting a conformational epitope preserved after paraformaldehyde fixation. Affinity-purified antibodies targeting this loop trace native UPK3A in umbrella cell apical membranes, ureteric epithelium, and cultured urothelial sheets, delivering crisp surface rings and single-band clarity on immunoblots with no cross-reaction to UPK1B or UPK2.
