Mouse Carnosine Synthase 1 (CARNS1) is a cytosolic ligase that catalyzes the ATP-dependent condensation of L-histidine and β-alanine to form the dipeptide carnosine. Highly expressed in skeletal muscle, heart, and olfactory bulb, CARNS1 maintains millimolar carnosine levels that buffer intracellular pH, quench reactive carbonyls, and modulate calcium sensitivity. Loss of CARNS1 reduces muscle carnosine, impairing contractile endurance and antioxidant defense. As the sole mammalian carnosine-synthesizing enzyme, CARNS1 is an important target for studies of muscle metabolism, aging, and carbonyl stress.
