Rat phosphodiesterase 4B (PDE4B) is an 85 kDa cAMP-specific hydrolase whose N-terminal targeting and UCR domains anchor the catalytic pocket to the ER and perinuclear membranes, lowering local cAMP to sharpen PKA signaling. A solvent-exposed 16-aa "KVE" loop (aa 495-510) between helices 15 and 16 within the catalytic site folds into a β-hairpin unique among rat PDE4 isoforms, presenting a conformational epitope preserved after fixation. Affinity-purified antibodies targeting this loop trace native PDE4B in cerebral cortex, striatal neurons, and cardiac myocytes, delivering crisp perinuclear rings and single-band clarity on immunoblots with no cross-reaction to PDE4A or PDE4D.
