Target Information
Mouse nectin-2 is a 60 kDa single-pass Ig-like adhesion molecule whose N-terminal V-set domain locks trans-homophilically or heterophilically to nectin-3 and DNAM-1, clustering at adherens junctions and synapses. A solvent-exposed 14-residue "QRD" loop (aa 80-93) within the V-domain folds into a β-hairpin unique among nectin paralogs, offering a fixation-stable epitope absent from nectin-1 or nectin-4. Affinity-purified antibodies targeting this loop trace native nectin-2 in Sertoli-cell tight junctions, hippocampal mossy fibers, and vascular endothelium, delivering crisp membrane doublets and single-band clarity on immunoblots with no cross-reaction to other nectin family members.
