Rabbit HLA-DR alpha is a 34 kDa type I membrane glycoprotein whose N-terminal β1 domain locks onto peptide side chains while an exposed DE-loop (aa 52-66) frames the antigen-binding groove, offering a species-unique epitope absent from mouse or human paralogs. A juxtamembrane cysteine pair stabilizes the DRβ contact surface, resisting fixation-induced denaturation. Affinity-purified antibodies raised against this loop trace native DRα on B-cell membranes, thymic epithelium, and splenic dendritic cells, delivering crisp surface staining and single-band specificity on immunoblots with no cross-reaction to DQ or DP isotypes.
