Rat T-cell immunoreceptor with Ig and ITIM domains (TIGIT) is a 26 kDa type-I transmembrane glycoprotein whose N-terminal IgV domain binds CD155 and CD112, while a C-tail ITIM recruits SHIP-1 to dampen NK and T-cell activation. A solvent-exposed 14-aa "SKP" loop (aa 52-65) between β-strands C and C' folds into a β-hairpin unique among rat Ig superfamily members, presenting a fixation-stable epitope absent from CD96 or DNAM-1. Affinity-purified antibodies targeting this loop trace native TIGIT in splenic NK cells, CD8+ T-cell follicles, and lamina propria lymphocytes, delivering crisp surface rings and single-band clarity on immunoblots with no cross-reaction to other coinhibitory receptors.
