Mouse peroxiredoxin 5 (PRDX5) is a 17 kDa atypical 2-Cys peroxiredoxin whose N-terminal peroxidatic cysteine reduces mitochondrial and cytosolic peroxides, while a C-terminal resolving cysteine regenerates the active site. A solvent-exposed 14-aa "KVE" loop (aa 77-90) between β6 and α4 folds into a β-hairpin unique among mouse PRDX isoforms, presenting a conformational epitope preserved after over-oxidation and fixation. Affinity-purified antibodies targeting this loop trace native PRDX5 in hepatic mitochondria, lung alveoli, and hippocampal neurons, delivering crisp punctate staining and single-band clarity on immunoblots with no cross-reaction to PRDX1 or PRDX3.
