Mouse Toll-Like Receptor Adaptor Molecule 1 (TICAM1, TRIF) is a cytoplasmic adaptor essential for TLR3 and TLR4 signal transduction. Upon ligand binding, TICAM1 recruits kinases and ubiquitin ligases, activating IRF3 and NF-κB to drive type-I interferon and pro-inflammatory cytokine production. Expressed in macrophages, dendritic cells, and fibroblasts, it bridges innate pathogen sensing to antiviral immunity. Loss of TICAM1 impairs host defense and alters inflammatory balance. As a critical node in TLR signaling, TICAM1 is an important target for studies of infection, inflammation, and vaccine adjuvanticity.
