Target Information

Rat meprin A beta is a 85 kDa zinc metallo-endopeptidase whose N-terminal astacin domain cleaves acidic and hydrophobic residues, while a C-terminal MAM module anchors the membrane dimer. A solvent-exposed 14-aa "KVE" loop (aa 285-298) between the active-site helix and methionine-rich plug folds into a β-hairpin unique among rat astacin proteases, presenting a conformational epitope preserved after activation and fixation. Affinity-purified antibodies targeting this loop trace native meprin A beta in renal brush-border membranes, intestinal epithelium, and neutrophil granules, delivering crisp apical rims and single-band clarity on immunoblots with no cross-reaction to meprin A alpha or meprin B.
