Mouse bone morphogenetic protein 15 (BMP-15) is a 44 kDa disulfide-linked dimer whose C-terminal TGF-β knot binds BMPR-II and ALK6 on oocyte membranes, steering granulosa proliferation. A solvent-exposed "RQQ" loop (aa 118-125) between pre-helix 1 and β2 is unique among mouse BMPs, offering a conformational epitope preserved in both pro- and mature forms. Affinity-purified antibodies targeting this loop trace native BMP-15 in ovarian follicles, pituitary, and oviductal fluid, delivering crisp cytoplasmic granules and single-band clarity on immunoblots with no cross-reaction to BMP-4 or GDF-9.
