Mouse eukaryotic translation initiation factor 3A (eIF3a) is a 166 kDa PCI-domain scaffold whose 12 structural repeats cradle the 40S ribosomal subunit, orchestrating start-codon selection. A solvent-exposed 18-aa "KVE" loop (aa 1,080-1,097) between PCI blades 8 and 9 folds into a β-hairpin unique among mouse eIF3 subunits, presenting a conformational epitope preserved after fixation. Affinity-purified antibodies targeting this loop trace native eIF3a in cytoplasmic mRNPs, neuronal dendrites, and proliferating fibroblasts, delivering crisp reticular staining and single-band clarity on immunoblots with no cross-reaction to other eIF3 subunits.
