Target Information

Mouse pronociceptin (PNOC) is a 19 kDa precursor whose C-terminal heptapeptide flanks nociceptin/orphanin-FQ, a ligand for the opioid-receptor-like-1 (NOP) receptor that dampens pain and stress signals. A solvent-exposed 14-aa "KVE" loop (aa 95-108) between the Arg-Arg cleavage site and the mature peptide folds into a β-hairpin unique among mouse opioid precursors, presenting a conformational epitope preserved after pro-hormone convertase processing and fixation. Affinity-purified antibodies targeting this loop trace native pronociceptin in hypothalamic perikarya, spinal dorsal horn, and adrenal medulla, delivering crisp cytoplasmic granules and single-band clarity on immunoblots with no cross-reaction to proenkephalin or prodynorphin.
