Mouse defensin alpha 5, Paneth cell-specific (Defa5) is a 4-kDa cationic antimicrobial peptide exclusively expressed in mouse small intestinal Paneth cells. It is stored in secretory granules and released into the intestinal lumen, exerting broad-spectrum activity against bacteria, fungi, and viruses by disrupting microbial membranes. Beyond direct antimicrobial defense, it modulates the gut microbiota composition and reinforces the intestinal epithelial barrier, critical for murine mucosal homeostasis. It also interacts with immune cells to regulate local inflammatory responses. Dysregulated expression associates with intestinal inflammation and dysbiosis. As a key mediator of gut mucosal defense and microbiota balance, mouse Defa5 is a critical target for studying intestinal biology, inflammatory bowel diseases, and host-microbe interactions.
