Target Information

Phospholipid hydroperoxide glutathione peroxidase (GPX4) is a crucial antioxidant enzyme that directly reduces phospholipid hydroperoxide, even when they are integrated into membranes and lipoproteins. Additionally, it can reduce fatty acid hydroperoxide, cholesterol hydroperoxide, and thymine hydroperoxide. The GPX4 gene is vital for mouse embryonic development, as its absence results in embryonic death between the embryonic stages E6.5 and E7.5 due to the embryo's inability to initiate development and form a blastocoel. Moreover, the lack of GPX4 disrupts overall embryonic development, leading to resorption during the embryonic period. In adult mice, GPX4 deficiency causes severe neurodegeneration. Research has shown that in the absence of GPX4, the mouse brain experiences significant neuronal death, particularly in the hippocampus, which is linked to neuronal apoptosis and astrocyte proliferation.
