Target Information
Tjp1, TjpP2, and Tjp3 are closely related scaffolding proteins that connect tight junction (TJ) transmembrane proteins, such as claudins, junctional adhesion molecules, and occludin, to the actin cytoskeleton. Tight junction protein 1 forms a multistranded TJP1/ZO1 condensate, which elongates to create a tight junction belt anchored at the apical cell membrane through interaction with PATJ. In the liver, TJP1 plays a key role in regulating insulin sensitivity and circadian rhythm. Research has demonstrated that TJP1 loss can enhance insulin sensitivity and improve liver metabolic function, including reducing blood sugar and insulin levels and ameliorating fatty liver's pathological characteristics. This indicates that TJP1 influences the liver's circadian rhythm via the mTOR signaling pathway, ultimately affecting insulin sensitivity. Additionally, TJP1 exhibits inhibitory effects in various tumors, particularly in renal clear cell carcinoma (KIRC). Studies show that TJP1 overexpression can suppress tumor cell proliferation and growth by inducing autophagy.
