Peptidyl-prolyl cis-trans isomerase B (PPIase B) is an enzyme that catalyzes the cis-trans isomerization of proline imidic peptide bonds in oligopeptides, potentially aiding in protein folding. In mice, CypB plays a wide range of roles, including in bone development, metabolic diseases, immune responses, inflammation, and tumorigenesis. While its PPIase activity is central to its function, interactions with other proteins contribute to its complex biological significance. CypB deficiency in mice can result in severe bone developmental abnormalities such as severe osteoporosis (severe osteopenia). Research indicates that CypB deficiency disrupts the normal folding of type I collagen, causing defects in bone matrix formation, which is linked to altered interactions between CRTAP and P3H1, key components in the collagen 3-hydroxyproline synthesis pathway.
