Target Information
Mouse Alpha-2-Macroglobulin-P (A2m) is a broad-spectrum protease inhibitor that plays a crucial role in various physiological processes, including inflammation, immunity, and tissue repair. A2m forms a tetrameric cage around active proteases, physically obstructing their interaction with substrates, a mechanism often referred to as the "protease snap-trap" or "venus-flytrap" mechanism. This protein also binds to microbial proteases, contributing to the host defense against infections. For instance, A2m can bind to the metalloprotease from Vibrio vulnificus and the cysteine protease cruzipain from Trypanosoma cruzi, enhancing the uptake and clearance of these pathogens by macrophages. Additionally, A2m has been shown to reduce apoptosis in infected cells and protect against oxidative stress, suggesting its role in maintaining cellular integrity during infections. In mouse models, A2m deficiency leads to increased susceptibility to infections and more severe inflammatory responses. Furthermore, A2m has been implicated in the regulation of lipid metabolism and cardiovascular health, highlighting its multifunctional role in maintaining homeostasis.
