Target Information
Rat tryptophanyl-tRNA synthetase (WARS1) is a 53 kDa class-I enzyme whose Rossmann fold grips ATP and tryptophan, while an appended KMSKS loop seals the active site during aminoacylation. A solvent-exposed 14-residue "HIGH" insertion (aa 45-58) within the catalytic cleft is unique among synthetases, presenting a conformational epitope preserved in both cytoplasmic and secreted fragments. Affinity-purified antibodies targeting this insertion trace native WARS1 in liver, lung, and activated macrophages, yielding crisp cytoplasmic puncta and single-band specificity on immunoblots with no cross-reaction to tyrosyl- or leucyl-tRNA synthetase.
