Target Information

Rat period circadian protein 2 (PER2) is a 125 kDa nucleocytoplasmic clock scaffold whose N-terminal PAS-A/B domains clasp CRY1 while its C-terminal CK1ε-binding pocket sets the pace of phosphorylation-driven degradation. A unique 40-residue "hypersensitive loop" (aa 700-740) emerges only in the non-phosphorylated, transcriptionally active conformation, offering a selective epitope absent from paralogs PER1 and PER3. Antibodies raised against this loop detect native PER2 in suprachiasmatic nucleus, liver sinusoids, and cultured fibroblasts, delivering crisp nuclear/cytoplasmic oscillations with single-band specificity on immunoblots and no cross-reactivity to phosphorylated or ubiquitinated forms.
