Target Information
Rat gastric intrinsic factor is a 55 kDa glycoprotein whose N-terminal α-helix grips vitamin B12 while a C-terminal β-sheet locks onto cubilin in the ileum, escorting the vitamin through digestion. A solvent-exposed 16-residue "QPK" loop (aa 120-135) between domains 2 and 3 folds into a β-hairpin unique among rat cobalamin carriers, presenting a conformational epitope preserved after paraffin fixation. Affinity-purified antibodies targeting this loop trace native intrinsic factor in gastric chief cells, pyloric glands, and duodenal lumen, delivering crisp apical granules and single-band clarity on immunoblots with no cross-reaction to cobalamin-binding proteins from serum or salivary glands.
