Porcine lipopolysaccharide binding protein (LBP) is a 62-kDa acute-phase glycoprotein primarily produced by porcine hepatocytes and secreted into the circulation, with additional expression in pulmonary and intestinal mucosal tissues. It specifically recognizes and binds lipopolysaccharide (LPS) from Gram-negative bacterial cell walls, forming stable complexes that facilitate LPS delivery to the CD14-TLR4-MD2 receptor complex on porcine monocytes, macrophages, and endothelial cells. This interaction amplifies innate immune activation-triggering pro-inflammatory cytokine release, phagocyte recruitment, and pathogen elimination-while balancing LPS detoxification to mitigate excessive tissue damage. Highly inducible during bacterial infections or tissue inflammation, it is a key mediator of porcine host defense against Gram-negative pathogens. As a critical link between pathogen sensing and immune response in swine, porcine LBP is a target for studying veterinary infectious diseases, sepsis, and mucosal immunity in livestock.
