Target Information
Mouse glutathione reductase is a 114 kDa homodimeric flavoprotein whose central funnel-shaped active site transfers electrons from NADPH to oxidized glutathione, regenerating cytosolic GSH pools. A solvent-exposed 14-aa "KVE" loop (aa 340-353) between the FAD and NADPH domains folds into a β-hairpin unique among mouse oxidoreductases, presenting a conformational epitope preserved after aldehyde fixation. Affinity-purified antibodies targeting this loop trace native GR in hepatic cytosol, erythrocyte membranes, and neuronal somata, delivering crisp diffuse staining and single-band clarity on immunoblots with no cross-reaction to thioredoxin reductase or glutathione peroxidase.
