Mouse Methylthioadenosine Phosphorylase (MTAP) is a cytoplasmic enzyme that salvages methylthioadenosine generated during polyamine biosynthesis, producing adenine and 5-methylthioribose-1-phosphate. Expressed ubiquitously, it conserves adenine nucleotides and links polyamine metabolism to purine pools. MTAP loss sensitizes cells to purine antimetabolites, making it a key metabolic marker. As an essential component of one-carbon and nucleotide pathways, MTAP is an important target for studies of metabolic stress, antimetabolite therapy, and tumor biology.
