Mouse DNA-Damage-Inducible Transcript 3 (DDIT3, CHOP) is a stress-inducible transcription factor that heterodimerizes with C/EBP and ATF proteins to regulate genes controlling apoptosis, amino-acid metabolism, and redox balance. Expressed at low levels under basal conditions, it is rapidly up-regulated in liver, pancreas, and macrophages by endoplasmic-reticulum stress, oxidative damage, or nutrient deprivation. DDIT3 translocates to the nucleus, where it represses anti-apoptotic and growth-promoting genes while activating pro-death signals. As a central executor of stress-induced cell death, DDIT3 is an important target for studies of unfolded-protein response, metabolic stress, and cytotoxic injury.
