Mouse citrate synthase (CS) is a 49-kDa homodimeric enzyme exclusively localized in the mitochondrial matrix of all mouse tissues, with highest expression in metabolically active organs (heart, skeletal muscle, liver). It catalyzes the rate-limiting step of the tricarboxylic acid (TCA) cycle, condensing acetyl-CoA and oxaloacetate to form citrate-linking glycolysis to mitochondrial energy production. By regulating acetyl-CoA flux into the TCA cycle, it controls ATP generation, fatty acid synthesis, and cellular redox homeostasis. Essential for murine energy metabolism in physiological states (e.g., development, exercise) and pathological contexts (e.g., metabolic disorders, mitochondrial dysfunction), it maintains tissue function through bioenergetic support. As a core TCA cycle enzyme and mitochondrial marker, mouse CS is a critical target for studying metabolic biology, mitochondrial diseases, and energy-related pathologies.
