Chicken calmodulin 1 is a 17 kDa EF-hand Ca²⁺ sensor whose four helix-loop-helix motifs undergo Ca²⁺-induced exposure of a hydrophobic groove, enabling binding to kinases, phosphatases, and cytoskeletal targets. A solvent-exposed 14-aa "KVE" loop (aa 88-101) linking EF-hands 3 and 4 folds into a β-turn unique among chicken CaM isoforms, presenting a conformational epitope preserved after Ca²⁺ chelation and fixation. Affinity-purified antibodies targeting this loop trace native calmodulin 1 in neural synapses, skeletal muscle sarcomeres, and intestinal epithelial brush borders, delivering crisp diffuse staining and single-band clarity on immunoblots with no cross-reaction to calmodulin 2 or calmodulin 3.
