Rat ADAMTS5 is a secreted metalloproteinase that degrades aggrecan and other cartilage proteoglycans. Expressed in joint cartilage and synovium, it cleaves the aggrecan interglobular domain, leading to cartilage breakdown and joint destruction. ADAMTS5 activity is up-regulated by inflammatory cytokines and down-regulated by mechanical loading. As a key enzyme in cartilage catabolism, ADAMTS5 is an important target for studies of osteoarthritis and joint repair.
