Mitochondrial uncoupling protein 2 functions as an antiporter, exporting dicarboxylate intermediates of the Krebs cycle in exchange for phosphate and a proton across the mitochondrial inner membrane, driven by mitochondrial motive force. This process influences glycolysis, glutaminolysis, and the glutathione-dependent redox balance. UCP2 plays a crucial role in the pancreatic β cells of mice, indirectly affecting insulin secretion by regulating mitochondrial oxidative phosphorylation and ATP production. Research indicates that the loss of UCP2 results in increased ATP levels in pancreatic β cells, inhibiting insulin secretion, while UCP2 overexpression enhances insulin secretion. Additionally, the absence of UCP2 is linked to insulin resistance and diabetes. For instance, in an obesity-induced diabetic mouse model, UCP2 expression is significantly upregulated, and its normalization can improve insulin secretion and blood glucose levels.
