Target Information
Suppressor of cytokine signaling 1 (SOCS1) serves as a crucial negative regulator of interferon (IFN) signaling types I and II, as well as other cytokines, including IL-2, IL-4, IL-6, and leukemia inhibitory factor (LIF). It attenuates cytokine signaling by inhibiting the JAK/STAT pathway, functioning by binding to JAK proteins and IFNGR1, thereby suppressing their kinase activity. SOCS1 is integral to the mouse immune system, especially in modulating T cell and macrophage activity. Mice lacking SOCS1 (SOCS1-/-) exhibit severe immune deficiencies, including lymphopenia, spleen and thymus dysplasia, and heightened sensitivity to several cytokines like IFN-γ, IL-2, IL-4, and IL-17. Additionally, SOCS1-/- mice are susceptible to severe inflammatory responses post-infection, leading to conditions such as hepatic steatosis, liver necrosis, and inflammation across multiple organs.
