Target Information
Aquaglyceroporins form homotetrameric transmembrane channels, with each monomer independently facilitating the transport of glycerol and water across the plasma membrane along their osmotic gradient. AQP9 primarily serves as the main transport channel for glycerol in the liver and plays a role in hepatic gluconeogenesis. Research indicates that AQP9 is the primary pathway for glycerol uptake in mouse hepatocytes, and its expression level is closely linked to hepatic gluconeogenesis. In mice lacking the AQP9 gene, hepatocyte glycerol uptake is significantly reduced, leading to diminished hepatic gluconeogenesis and impacted blood sugar levels. Furthermore, AQP9 expression is regulated by insulin and leptin; in mice with insulin or leptin deficiencies (such as ob/ob mice), AQP9 expression is elevated, potentially related to a high-fat diet and insulin resistance.
