Target Information
E3 ubiquitin-protein ligase CHIP is an enzyme that directs misfolded chaperone substrates towards proteasomal degradation. It is involved in maintaining mitochondrial morphology and facilitating the mitophagic clearance of dysfunctional mitochondria, thus serving as a defender against apoptosis due to cellular stress. A deficiency in STUB1 results in the accumulation of unfolded proteins, accelerating cellular aging and reducing lifespan. For instance, mice lacking STUB1 show signs of accelerated aging, including cognitive dysfunction, motor impairment, and a shortened lifespan. Additionally, STUB1 plays a role in regulating the ubiquitination of the core circadian protein BMAL1, thereby contributing to the inhibition of cellular aging.
