Target Information
NADH dehydrogenase [quinone] 1 is a flavin-containing quinone reductase that facilitates the two-electron reduction of quinones to hydroquinones, utilizing either NADH or NADPH as electron donors. Through a ping-pong kinetic mechanism, electrons are transferred sequentially from NAD§H to the flavin cofactor and subsequently from the reduced flavin to the quinone, thereby avoiding the formation of semiquinone and reactive oxygen species. NQO1 acts as a crucial antioxidant enzyme by catalyzing the two-electron reduction of quinone compounds, which helps protect cells from oxidative stress and damage associated with the redox cycle. For instance, NQO1 knockout mice (NQO1−/−) exhibit heightened sensitivity to menadione toxicity, underscoring NQO1's essential role in defending against oxidative stress.
